That was my first thought too. I would look at the fstab file and see how the volume is mounted. You may find it is declared RO and need to make it RW.
Can't say I have done this before with a USB. One concern is the large wear effect from writes.

I've never done what you are asking for, but my first thought would be
to simply treat the USB device as a regular disk. Whatever you come
up with, please share it with the group, I'm at least one person
interested in the outcome.
